I have a 2021 Zion Slumber with a 400 amp lithium battery system. I have winterized the van and it is not in use for the winter. How long should the batteries hold a charge? Do I need to plug into an AC outlet?

No one can answer that without knowing the demands on the battery. I suggest checking the battery maybe weekly at first, see how it's holding, then consider longer durations between checks. If there are no demands or parasitic loads it should last a long time.
 
You mentioned you have the rig winterized. Are you in a cold climate? If so you have to worry more about low temps harming the batteries. If there are no parasitic draws on the battery it should hold a charge for a long time.
