I purchase an 1993 Ford E350 Conquest 101 with a 460 motor.

Purchase it but did not start.

Didn’t have fuel pressure after replacing fuel pump, filter and fuel regulator in which I replace return fuel line to tank and I now have low fuel pressure.

When I kink the return hose I get pressure and does not leak down but if not kinked it will leak pressure rapidly.

Anyone have any ideas on what to look at next?

As mentioned above - either a leaky fuel regulator or a leaky back flow valve at the fuel pump.

Will it briefly start when a small amount of starting fluid / ether / "explosion in a can" is sprayed into the carburetor or throttle body? I don't like using starting fluid as a crutch to get an engine to start because its not good for the oil film between the pistons and cylinder walls. But its a heck of a diagnostic tool when used sparingly.

Put in new fuel pump, fuel filter, new return line (which had no fuel pressure before return line).
Will not start, barely will fire but fuel pressure drops rapidly.

You said you kink return line you have pressure. Bad regulator!!!
Don't care if you replaced it. When kinked is pressure as high as it should be? Yes, put another regulator in.
